Treatment failure with antidepressants and off-label treatment with minocycline for major depression: 4 case reports In an open pilot trial, 4 patients [ages and sexes not stated] were described, who experienced treatment failure following treatment with citalopram, escitalopram, vilazodone or desvenlafaxine for major depression. Additionally, the patients received off-label treatment with minocycline for major depression [routes not stated]. The patients, who had major depression, had been treated for at least 6 weeks with stable dosages of citalopram 20 mg/day (1 patient), escitalopram 20 mg/day (1 patient), vilazodone 20 mg/day (1 patient) and desvenlafaxine 50 mg/day (1 patient) during the study trial. However, the patients failed to achieve substantial improvement in the depressive symptoms after at least 6 weeks of the treatment. Therefore, they received off-label minocycline 100mg twice daily for 8 weeks as an augmentation therapy along with their existing treatment regimen. At the end of 8 weeks of treatment with minocycline, the patients achieved remission in the major depression. Avari JN, et al.
