Abstract
Hematopoietic stem cell collection (HPSC) techniques contribute immensely to the outcome of allogeneic and autologous transplants. The major sources of hematopoietic stem cells are the bone marrow, cord blood at birth, or peripheral blood. The technique of collection of HPSC determines the outcome of allogenic and autologous transplants. The techniques adopted should ensure adequate CD34 cell dose with minimal number of collection procedures. Cord blood requires a single,

adequate, aseptic collection technique, using cord blood collection kits, at birth, in a wellmonitored pregnancy. Allogeneic bone marrow is collected without administration of G-CSF. Allogeneic peripheral blood stem cells (PBSC) require G-CSF mobilization, whereas autologous PBSC collection will require G-CSF alone, G-CSF and chemotherapy, and G-CSF and upfront use of plerixafor to ensure adequate collection of CD34 cell dose to serve dual purpose of proceeding with the transplant and also with a prospect of cryopreserving a suitable cell dose for a possible future tandem transplant. Technological advances and intuitive software incorporated in modern-day cell separators enable PBSC apheresis collection procedures to be done safely with predictable
