Amiodarone associated with low AE rate Although amiodarone increases rates of heart, liver, lung, neurological, skin and thyroid disorders compared with placebo, it appears to be associated with a low overall adverse event (AE) rate according to findings of a meta-analysis published in the American Journal of Cardiology. ClinicalTrials.gov, the Cochrane Central Register, Google Scholar and PubMed were searched for randomised placebo-controlled trials investigating the use of amiodarone; 43 trials in a total of 11 395 patients met inclusion criteria for a meta-analysis evaluating incident rates of AEs per 10 000 patient-years (IRs) and the relative risk (RR) of AEs with amiodarone treatment versus placebo. The overall risk of AEs was higher in amiodarone recipients than in placebo recipients (IR 129 vs 74; RR 1.77; p=0.002).
