Abstraet-A method of i~)Iation of bioactive ,a.ater-soluble saponin-free subti'action (RG-ws-l-l-l) li'om Korean red ginseng using solvent extraction, ultrafiltration and chromatography on Diaion HP-20 resin was developed. 3b e',,aluate the potency of RC~-ws-l-l-t to effect on the heat stress (HS) response and antMnflammatory response in ncutrophils, the oxygen consumption (OC) and Ibrmation of superoxide radicals (FSRt ~.ere assayed in suspensions of neutrophils li'om mice. Supplementation of RG-ws-l-l-I to mice in rh'o (200 mg/kg, 14 days intrapcritoneally) results in the increase OC by' neutrophils ass~,ed ex vivo at 37 "C. We proposed to use the functional model system, namely murine neutrophils with regard to their physiological functions. This cell system is quite suitable to evaluate anti-inflammatory and anti-heat stress effects of Korean red ginseng (KRG) supplemented either in vn,o or in vitro, In fact neutrophils are responsible for defense against pathogens. Neutrophils are the primary cells that are involved in the killing of bacteria via an oxygen-dependent anti-microbial system, thal is able to form strong oxidants - reactive oxygen species including superoxide ion (O:), hydrogen peroxide (H_,O,), hypochlorous acid (HOCI) and hydroxyl radical (Oil"), When nentmphils are exposed to microo~anisms, an increase in the rate of ~ consumption (the respiratory burst) is induced. An increased formation of reactive oxygen species is the consequence and strong oxidants like hypochlorous acid and H_~O_~are released into the intracellular space of neutrophik [Badwey and Kamovsky, 1980].
