Mesothelioma remains a lethal cancer with increasing global incidence. While there have been modest incremental advances, there is no clear optimal surgical approach, and presently, only one standard systemic treatment option, cisplatin and pemetrexed, is widely approved. Median overall survival remains, even with aggressive multimodality treatment, poor. Many attempts over the past decade to improve upon the established chemotherapy regimen and surgical treatment paradigm have been unsuccessful. While there has been an influx of new systemic treatments over the past 10 years for patients with molecularly characterized malignancies, no such progress has been made for patients with malignant mesothelioma. Similarly, the advent of immunotherapy with checkpoint inhibitors is under investigation in mesothelioma. These agents do appear active but, as in other diseases, only in a subset of patients. Consequently, a tremendous unmet need remains for mesothelioma treatments, and much research is ongoing. Within this book, Mary Hesdorffer and Gleneara E.
