Abstract This present study aims to develop a liquid chromatography coupled to tandem mass spectrometry assay to quantify phillyrin in rat plasma and tissues. Plasma and tissue samples were processed via protein precipitation, and calceolarioside B was chosen as the internal standard. Selected reactions monitoring mode was applied to detect the analytes of all samples. Mass spectrometry was conducted in the negative ion mode for detection. The MS/MS ion transitions monitored were m/z 533.4 → 370.8 for phillyrin and m/z 477.3 → 161.0 for calceolarioside B, respectively. All calibration curves were linear over the range of 1– 800 ng/ml in each biological matrix. The lower limit of quantification for plasma and tissue homogenates was 1 ng/ml. Inter- and intra-day precision and accuracy well met within the acceptable ranges (≤ 15%). It was successfully applied to the pharmacokinetic and tissue distribution studies of phillyrin after intravenous doses of 0.1 mg/kg in rats. Keywords Pharmacokinetics . Tissue distribution . Introduction Forsythia suspensa (Thunb.) Vahl, Oleaceae, also known as “Lian Qiao,” is a famous traditional Chinese medicine used to treat infectious diseases, such as gonorrhea, ulcer, pyrexia, erysipelas, and inflammation (Li and Chen 2005; Han et al. 2017). As an alternative of its fruits, its leaves have been consumed as medicinal beverages in China due to their versatile health benefits (Qu et al. 2008a). The Mahuang Lianqiao Chixiaodou decoction containing “Lian Qiao” is used to treat children with acute glomerular nephritis by effectively improving renal function, relieving inflammatory reaction, and alleviating local pathological and physiological states (Zheng and Zhuo 2006; An and Bai 2019). Many pharmacological studies proved that phillyrin displays various biological activities, such as antioxidant, anti-inflammatory, anti-hyperlipidemic, and antipyretic activities (Zhao et al. 2005; Qu et al. 2008b; Zhong et al. 2013; Kong et al. 2014; Jiang et al. 2020). It also lowers the body weight in obese mice by modulating the PPAR/-angiopoietin-like protein 4 pathway (Xiao et al. 2018).
