Abstract The role of lncRNA LEF1-AS1 in the regulation of osteogenic differentiation of dental pulp stem cells (DPSCs) is still obscure. Here, we demonstrated that LncRNA LEF1-AS1 expression was associated with osteogenic differentiation of DPSCs and overexpression of LEF1-AS1 promoted osteogenic differentiation. Moreover, lncRNA LEF1-AS1 and miR24-3p could directly regulate each other and LEF1-AS1 acted as sponge partner of miR-24-3p. Furthermore, LEF1-AS1 and miR-24-3p synergized to regulate osteogenic differentiation of DPSCs. Finally, we verified TGFBR1 was the direct target of miR-24-3p in osteogenic differentiation of DPSCs and miR-24-3p/LEF1-AS1 sponged to regulate TGFBR1 expression. Our study revealed a novel mechanism about how did lncRNA LEF1-AS1 execute function in osteogenesis of DPSCs and thus might serve as potential therapeutic target for the bone regeneration in the dental pulp. Introduction The pulp tissue is located in the pulp cavity inside the tooth, which is the only soft tissue in the tooth [1, 2]. Fibroblasts isolated from Pulp tissue are called dental pulp stem cells (DPSCs) [3]. Dental pulp stem cells have multidirectional differentiation potential [4]. In addition to cells capable of forming mineralized nodules, they can also differentiate into fat, bone, cartilage, muscle, vascular endothelium, liver, nerve and other cell lines after induction of different cytokines [5–7]. More than 14 million root canal operations are performed each year to treat bacterial pulpitis, characterized by inflamed pulp and periapical tissues [8]. The occurrence of pulpitis will lead to the loss of regeneration ability of dental pulp cells, making the complete healing of pulpitis Yuexia Wu and Keqian Lian are contributed equally. * Cong Sun [email protected] 1

difficult [9]. Thus, the discovery of effective treatment targets for pulpitis with little side effects is quite essential. To date, knowledge about the regulation of DPSCs differentiation still remained vague. Long non-coding RNAs (lncRNAs) are RNAs with a length of more than 200 nucleotides and are not translated into proteins [10–12]. Their functional disorders and specific roles in various diseases, especially cancers, have attracted increasing attention [13, 14]. Alterations or mutations in the lncRNAs have been reported to modulate multiple physiological processes [14]. LEF1-AS1 (LEF1 antisense RNA 1), encoded in the lymphoid enhancer‐binding
