Depressed mood disorders with anastrozole A recent communication from the Netherlands Pharmacovigilance Centre Lareb notes an association between anastrozole treatment and depressed mood disorders. The non-steroid aromatase inhibitor is indicated for postmenopausal women with hormone-receptor-positive breast cancer. The mechanism of action, resulting in lowered serum estradiol levels, could be associated with mood disturbances. Depression has been associated with both letrozole and exemestane, which may suggest a class effect. The centre has received 14 reports of depressed mood in anastrozole recipients, comprising depressed mood (n=8), depression (n=5) or depressive symptom (n=1), occurring 1 day to 10 months after treatment initiation. Anastrozole was withdrawn in 10 patients, while the dose was not changed in 4 patients. Similar associations of anastrozole and depressed mood disorders are found in the Eudravigilance and Vigibase databases. Mood disturbances are described in the Dutch Summary of Product Characteristics, but the Patient Information Leaflet does not mention mood disturbances or similar events as adverse drug reactions.
