Antigen-Specific Activation of Blood Lymphocytes in Mice Immunized with Brucella abortus 19 BA

Balakhonov Translated from Byulleten’ Eksperimental’noi Biologii i Meditsiny, Vol. 169, No. 4, pp. 468-471, April, 2020 Original article submitted November 29, 2019 We studied the expression of activation markers CD25 and CD69 by blood lymphocytes in white mice vaccinated with Brucella abortus 19 BA in antigen-specific tests in vitro. During incubation of blood lymphocytes with brucellosis polysaccharide-protein antigen, a statistically significant increase in the expression of CD25 by B cells and CD69 by T cells was observed; brucellin increased the expression of CD25 by B and T cells. Comparative analysis of the action of antigen preparations B. abortus showed that only brucellin has antigen-specific activity against CD19+CD25+ cells. The used method can be considered as a promising test for evaluation of the effectiveness of brucellosis immunoprophylaxis, which substantiates the need for further research. Key Words: brucellosis; lymphocyte activation; flow cytometry; antigens Brucellosis is an acute invasive infectious disease with a high potential for chronification. As the epidemiological situation for brucellosis in the Russian Federation remains unstable and challenging due to unfavorable epizootic situation, vaccination/revaccination of people living in endemic areas is conducted annually [2]. For specific prevention of this disease in humans, a live vaccine based on Brucella abortus 19 BA strain is used. Currently, assessment of the immune status of subjects vaccinated/revaccinated against brucellosis consists in the Burnet skin test and serological tests that have a number of disadvantages. In particular, Burnet test can cause local and systemic pathological reactions, while serological tests often show false negative results [5,6]. The absence of regulations for the assessment of immunological effectiveness of vaccination against brucellosis dictates the need for improvement of methods with the use of modern analytical equipment.
