Olanzapine Fatal pulmonary embolism: 6 case reports

Six patients developed fatal pulmonary embolism during treatment with olanzapine [dosage and duration of treatment before reaction onset not stated for any patients; therapeutic indication not stated for five patients]. Characteristics for the six patients are described in Table 1. Of the six patients, a 47-year-old woman (patient 1), who had schizophrenia, a body mass index of 19 and was treated with lithium and, most recently olanzapine, was found dead. Her autopsy was remarkable for acute pulmonary embolism in the left main pulmonary artery which extended into the lobar branches; an adherent thrombus in the left popliteal vein was revealed when leg vessels were dissected. The remainder of the woman’s autopsy revealed there no evidence of natural disease or trauma. The presence of olanzapine was confirmed by toxicology, but no other substances were found; there were no other risk factors for pulmonary embolism. Two of the other six patients had risk factors for pulmonary embolism. Factor V Leiden and prothrombin mutation testing was negative all in patients.
